Amazon or Matador (2) Conversion software

Following my previous post, could anyone tell me what good/bad experiences they have had from free downloadable software for e-book/Kindle publishing.

I use Word/Windows, and of course whenever my anthology is 'finished' then I will be my own prof-reader, editor etc.I have seen brief articles about

(i) Calibre

(ii) Scribus

(iii) Mobipocket

Which is the best?

Does Amazon offer Free conversion software?

Do any major retailers offer the same?

I hear PC World does.

If you're publishing to Kindle and Smashwords you won't need software - you load your formatted Word document directly and they convert.

I've heard good things about Calibre but not tried it. No idea about the others.
