I just wondered, when sending off my three chapters to an agent, does the prologue count as a chapter or do I send the prologue and first three chapters?

This depends on your chapter lengths. Someone brought it up a few weeks back and the general concensus (between me and Louise) was to send whichever number of chapters landed you between 30 and 50 pages sent. So, if your prologue and first chapter takes you over thirty pages but your second would take you over fifty, you just send the prologue and chapter one. If your prologue and first three chapters all fit within fifty pages, send them all.
Of course, you should always check the individual guidelines of the literary agent in question to make sure they don't have a different suggestion.
